Is a 32-20 chambering reamer for a revolver cylinder different than one for a rifle?

No they are not the same. Larger pilot on the chamber reamer of the revolver and the lead is in the barrel in the barrel not in the cylinder.

If you buy a 32-20 reamer make sure you get a reamer drawing so you know what you're getting. I bought a Clymer barrel reamer from Brownells that had virtually no leade and no freebore. Accuracy was terrible. I had to order a custom ground throater to complete the chamber so it would shoot accurately. As mentioned in another thread, give Manson a call if you're thinking of buying one.
