Yes, that is the prefered method, with a tapered barrel. You see that a lot with bolt action rifles. That's so the top of the blocks and therefore the dove tails, are inline with the bore axis. But, if you can't obtain that, always make the front block lower and the closest match to the rear block.
That will generally put the rear mount, somewhere in it's midle range of adjustment, for our velocity range.
If you want the maximun adjustment and your closest range is 100 yards. Sight it in about 3" low at 100, with the lowest velociy load that you will shoot and keep raising the rear block, to maintain that "O".
If you have a way of measuring the distance between the bottom of the scope tube and the bottom of the cradle that it fits in, in the mount, that should be the amount that you can raise the rear block but, keep in mind that the bell of the scope, may interfere with your plans and you may have to raise the front block first.
