Am I correct in assuming that Remington never made the Number One Rolling Block in .40-65?

I think you're correct. At least I've never seen one, and Marcot's book doesn't show that caliber.
Since the .40-65 was introduced around the time Remington went bankrupt in 1886 it's a bit newer cartridge. So if it was ever offered I'd expect it to be a special order chambering. I'd never say "never", but it's not listed in the old catalogs of the 1890's nor as a cartridge available in the #5 Sporting Rifles either.
I have a Rolling Block I built off an original Sporting action and chambered in .40-65 Win. and it's one of my favorites to shoot.

Since you're back I'll ask you a question thats been nagging me.  Do the Sporting No. 1 and 1 1/2 actions share the same barrel threads?   I'm thinking yes.

Yellowhouse, They are not the same, at least I have never seen a 1-1/2 with the larger shank. The 1-1/2 has a thinner reciever thickneness. The 1-1/2 thread is very similar to the #2 but the configuration of the shank length and pilot stub are not the same.

They are both 12 square threads per inch, but a #1 is a 1" diameter, while #1 1/2 is .850" diameter.

I have a nice Rolling Block that a friend had built. I bought it from his daughter when he passed. My granddaughter likes it too.

That's a good looking Rolling Block. The recoil pad makes it a lot more comfortable to shoot I bet!
