Got to thinking about that sight.  Might it have been a shotgun originally?

Many have been converted to rifles.

Rolling Block shotguns I've seen had a grooved top receiver Joe to get low enough to see the bead front sight. I think this taller rear sight on the receiver would need a very tall bead to match up front.

  They are not grape leaves, at least not like any grape leaf I've seen, and I've seen lots. Maybe hops. I know very little about hops except that they are labor intensive.

I see both grape leaf and acanthus mixed into the stems. it's appealing, but may not be a normal rendering. It sure wouldn't be on Colts, which tended to have everything, but the leaves were usually done in a single style. Of course, never say never when dealing with firearms- and certainly with Colt.
As a guess, this one was to pass a test, maybe for a guild, and the craftsman included each to show his versatility.  He passed.
