A chamber on a 44 1/2 has a very tight neck. Could it have been designed for breech seating the bullet rather than loading it in the case?

Just the opposite ! A chamber intended for shooting breach seated cast lead bullets would have a tapered throat cut forward of the chamber end with a so called "throating reamer". Some such throats are cut longer than others. This work is not for a novice to accomplish. Questions ?

Yes. Many old rifles have very small chamber necks. I've found they shoot very accurate when the bullet is breach seated. Fixed ammo with the bullet being bore diameter works okay when using black powder loads. I think that was Stevens intent.
Many old rifles were chambered for bore diameter bullets.

Muzzle load/ breech seat were made that way. You will not breech seat without a mechanical leverage seater.

Yes you can. Use soft, 30-1, 20-1 tapered bullets that measure close to groove diameter at the base.
I do it all the time.

I have a .32-40 reamer that transitions from the neck diameter to bore diameter with a 45 degree angle. Can a chamber cut thusly be successfully used for breech loading?

99% of all cartridge rifles are chambered that way. All are breech loaders.
If you mean "breech seated", yes.

Yes, CPA Rifles has reamers that cut the leade for both breech & fix cartridge - call them

Gentlemen, thanks for the input. I have made a mechanical breech seater, but my bullets are .323 and too hard for it to work well. I think I will just turn the neck on a few brass and seat the bullets in the case. I am not quite ready to increase the neck diameter in the chamber, but suppose I am headed in that direction!

Sounds like your bullet is just to big so don't give up and work with what you have. You do not have to have a leade with a long tapered throat with some freebore as some might lead you to believe to make it work just a bullet that fits.

Yes, it can shoot very accurately (I had one rifle like that) but, you need a good fitting, tapered bullet. In that type of throat, a over sized bullet will take so much pressure to seat, that it will deform it. By over size, I mean a tapered bullet that has a a base band .002/.003 larger than the groove dia. and the first bands only .001/.002 under groove dia. So, you may have a bullet with as much as .005 taper but actually only .001/.002 of actual taper and your swaging the rest of it into the barrel and it can deform in that process. Same goes for straight cylindrical bullets.
The most important part of the bullet is the seal on the base band so, it has to be at least .0005/.001 larger than the groove with your type throat.
Assuming a .315 bore and .321 groove, I would want a mold that casts the first band at .318 (no smaller than .317) and the base band at .322 with a fairly even taper in between.
BS it with .040 +- .010 left un-engraved on the base band and you'll have a good start to work up a good powder charge.
