They are set to correspond roughly to grains of black powder. If you are using smokeless You will need to try charges and weigh them to determine what they correspond to in weight of your particular powder.

With BP charges of 20 g. or more, I've found the slide graduations so close to what they should be that I don't even bother to test-weigh them anymore.

With your powder measure should be a card or sheet that shows how much smokeless of various types per mark on the drum.  The marks are, of course, for black powder.  So for blackpowder, I use them direct.  In my little booklet that came with my #55, were all of those wonderful powders that we lost due to ww ll.  Even Sharpshooter, very accurate, but hard on barrels.
